We investigate the effects of and associations with estrogen-based hormone replacement therapy (HRT) and muscle characteristics and body composition in postmenopausal women in order to explore estrogen and other steroid hormones as molecular mechanism for aging-induced muscle weakness. Especially the muscle-fat cross-talk will be in focus.<\/p>\n<p>Study subjects include both pre- and postmenopausal women as well as identical postmenopausal twin pairs discordant for HRT i.e. the other twin uses HRT for several years while the co-twin has never used HRT. Body composition, muscle-related phenotype data, as well as muscle, adipose tissue and blood samples (whole blood, serum, plasma, leukocytes) \u00a0\u00a0are available from the women. In addition to conventional cell and molecular biological and biochemical analyses also high through-put techniques like gene expression arrays, miRNA arrays, epigenomic, proteomic and metabonomic analyses are done or ongoing.<\/p>\n<p>Research collaboration within this study entity includes Finnish researchers at Universities Helsinki, Oulu, Kuopio and Jyv\u00e4skyl\u00e4 and internationally at Universities Uppsala, Padova, Ancona and Paris via MYOAGE and ENGAGE research consortia (EU\/FP7). The HRT specific knowledge, both in relation to sarcopenia and other health outcomes will add our understanding, when evaluating the possible positive and negative effects of HRT. This study will increase our knowledge of the genetic and molecular mechanisms behind sarcopenia and responsiveness to interventions upon aging. Erityinen tutkittavien ryhm\u00e4 on vaihdevuosi-i\u00e4n ohittanut 54\u201362 vuotiaiden identtisten kaksossiskojen ryhm\u00e4, jossa sisaret ovat HRT:n suhteen diskordantteja. Kaikille tutkittaville on tehty laajat lihasten ja kehon koostumuksen ja lihasten toimintakyvyn mittaukset sek\u00e4 terveydentilan, ravitsemuksen ja liikunta-aktiivisuuden kyselyt. Tutkittavilta on otettu lihas- ja rasvakudosn\u00e4ytteet sek\u00e4 verin\u00e4ytteet (koko veri, seerumi, plasma ja valkosolut). Tutkimusn\u00e4ytteist\u00e4 on tehty\/tehd\u00e4\u00e4n perinteisten solu- ja molekyylibiologisten ja biokemiallisten analyysien lis\u00e4ksi laajat koko perim\u00e4n kattavat geeniekspressio-, mikroRNA- ja DNA:n metylaatioanalyysit sek\u00e4 proteomiikka ja metabonomiikka-analyysit.<\/p>\r\n<p style=\"text-align: left;\">Tutkimusta tehd\u00e4\u00e4n yhteisty\u00f6ss\u00e4 Helsingin, Oulun, Kuopion ja Jyv\u00e4skyl\u00e4n yliopistoissa ty\u00f6skentelevien tutkimusryhmien kanssa. We investigate the effects of and associations with estrogen-based hormone replacement therapy (HRT) and muscle characteristics and body composition in postmenopausal women in order to explore estrogen and other steroid hormones as molecular mechanism for aging-induced muscle weakness. Especially the muscle-fat cross-talk will be in focus.\r\n\r\nStudy subjects include both pre- and postmenopausal women as well as identical postmenopausal twin pairs discordant for HRT i.e. the other twin uses HRT for several years while the co-twin has never used HRT. Body composition, muscle-related phenotype data, as well as muscle, adipose tissue and blood samples (whole blood, serum, plasma, leukocytes) \u00a0\u00a0are available from the women. In addition to conventional cell and molecular biological and biochemical analyses also high through-put techniques like gene expression arrays, miRNA arrays, epigenomic, proteomic and metabonomic analyses are done or ongoing.\r\n\r\nResearch collaboration within this study entity includes Finnish researchers at Universities Helsinki, Oulu, Kuopio and Jyv\u00e4skyl\u00e4 and internationally at Universities Uppsala, Padova, Ancona and Paris via MYOAGE and ENGAGE research consortia (EU\/FP7). The HRT specific knowledge, both in relation to sarcopenia and other health outcomes will add our understanding, when evaluating the possible positive and negative effects of HRT. This study will increase our knowledge of the genetic and molecular mechanisms behind sarcopenia and responsiveness to interventions upon aging.
